Activity 6 - An additional model of heterochromatin establishment and maintenance - Open

Authors have introduced Ura4 reporters to evaluate heterochromatic silencing at different regions (arrows in the figure).

They have inserted reporters in different mutant Yeast strains, each of them containing K.O. of one gene in the RNA interference pathway, i.e. dcr1 (dicer), rdp1 (rna-dependent polymerase), ago1 (argonaute1). Expression of Ura4 reporter was then examined by RT-PCR, as compared to another gene placed to euchromatic region (ura4 DS/E). wt means the wild-type yeast strain.


Based on what you see in this Figure, do you think that the RNA interference mechanism has any role in heterocrhomatin formation at centromeres in S. pombe ?
